MyoD and Mash1 of unspecified species both activated reporter gene expression when separately transfected into C3H10T1/2 mouse fibroblasts in addition to a luciferase reporter vector (six tandem repeats of putative Ascl3-E47 binding site "oligo A" -MEF-1 sequences-SV40 promoter-luciferase). Reviewer: Stuart Lithwick Ascl3 may be a transcription factor, which regulates gene expression. Ascl3 interacts with an oligonucleotide selected using CASTing as demonstrated by EMSA assay. However, competition assays were not included, lessening the impact of this data. In addition, Ascl3 reduces reporter gene expression when cotransfected into mouse fibroblasts with expression vectors for MyoD and Mash1 as well as a reporter vector construct. In the absence of Ascl3, the cotransfection of expression vectors for either MyoD or Mash1 with the reporter vector construct leads to increased reporter expression. |
