Reviewer: Stuart Lithwick Human Arid4b (referred to as SAP180 in this PMID) is likely a transcriptional cofactor, which represses gene expression in a complex with a variety of other co-repressor proteins. Arid4b represses luciferase reporter expression when fused to the GAL4-DBD and cotransfected into HEK-293 cells along with a luciferase reporter vector. In addition, Arid4b interacts with each of the components of the mSIN3A corepressor complex as demonstrated by co-immunoprecipitation from K562 nuclear extracts. This suggests that transcriptional repression by Arid4b is achieved via the action of Histone Deacetylase (HDAC). However, the addition of HDAC inhibitor TSA to the cotransfection experiment did not impact upon the ability of Arid4b to repress expression suggesting that it may be by other means. |
