In a co-immunoprecipitation assay to test whether zipzap/p200 protein binds to SRF in the presence of other SRF cofactor(s), zipzap/p200 was found in a protein complex with SRF and other SRF-binding proteins (including p49/STRAP and Nkx2.5) . Zipzap/p200 with SRF activated the promoter activity of cardiac actin, MLC2v, and ANF genes. Reviewer: Amy Ticoll The zipzap/p200 (Arid2) protein has two classic C2H2 zinc fingers. Zipzap/p200 co-immunoprecipitates with SRF and SRF cofactors, however a mammalian two-hybrid system suggests only a weak interaction between SRF and zipzap. Zipzap/p200 with SRF activated the promoter activity of cardiac actin, MLC2v, and ANF genes. Zipzap/p200 co-transfected with another SRF cofactor, myocardin, results in synergistic activation of the ANF promoter. |
