Reviewer: Amy Ticoll The mammalian SWI/SNF-related complexes facilitate gene transcription by remodeling chromatin using the energy of ATP hydrolysis, and may interact with site-specific transcription factors or other cofactors. Mass spectrometric analysis and immunoblotting demonstrate the presence of ARID1B in SWI/SNF-related complexes. Co-precipitation studies indicate that ARID1B associates with BRG1 and hBRM, essential ATPase subunits of SWI/SNF-related complexes. Analysis of DNA-binding behaviour in a in a DNA pull-down assay indicates that ARID1B binds DNA in a non-sequence-specific manner. ARID1B stimulates transcription mediated by the steroid hormone receptors. |
